Is Florida ready to meet #thebumblingbiochemist? And are you ready to meet some of the MOLECULAR FRIENDS I’ll be learning more about with some organismal friends (fellow scientists) at #EB2019? Today I arrived in Orlando for the 2019 annual meeting of the @ASBMB (American Society for Biochemistry and Molecular Biology) - part of a larger Experimental Biology conference. I’m excited to meet lots of awesome scientists (including @queening_in_science!). I’m not sure what I will be posting about the next few days so I thought it would be a good idea to re-introduce you to some of my molecular friends. _ All the awesome stuff I study - everything from DNA to proteins - and even the bench I work at - it’s all made up of ATOMS. These are made up of smaller parts - subatomic particles - there are positively charged parts called protons and the number of these an atom has determines its identity - we name ELEMENTS based on their number of protons. So if an atom has 8 protons we call it oxygen, 6 and it’s carbon, etc. http://bit.ly/2G8SYax _ Hanging out with the protons at the center of the atom (nucleus) are neutrons. These aren’t charged and there can be different numbers of these but usually something close to the number of protons. We call atoms of the same element (same number of protons) with different numbers of neutrons ISOTOPES. _ Some of these are unstable and let off radiation when they reorganize themselves to a more stable form. We can use such radioactive isotopes to label molecules to track them. They act the same as the “normal” form so they’re “minimally invasive” unlike things like fluorescent tags that can change the molecule’s natural properties. And we can detect tiny quantities so we can get really sensitive read-outs, but they do require special precautions to work with, so experiments take way longer than usual and it can be stressful to work with but so useful! http://bit.ly/2VtYSG7 _ Whizzing around the clump of neutrons and protons we call the nucleus are electrons. These are negatively charged and atoms can share them. When they do, strong covalent bonds are formed. PART 2

⚠️ New Wikipedia article alert!!!! ⚠️ Dr. NAMANDJE BUMPUS studies anti-HIV drug metabolism - metabowhatta? Metabolism is just a term we use to describe the chemical changes our body makes to molecules, and metabolite’s what we call the changed version. When you ingest a chemical that’s in one form, that’s not necessarily the form it stays in as it travels throughout your body and “does its thing.” You take some drug, your liver does something to it and, bam - it’s a metabolite. _ Just like the food you eat gets processed, the pharmaceutical drugs you take do too. They’re already pretty small (much smaller than that bite of sandwich) but they can still get broken down further. And they don’t just get broken down, they can also get added onto or just tweaked a bit. All of these changes fall under “metabolism” and it’s a big focus for pharmacologists, because these changes can affect the drugs’ activity - either positively or negatively. _ A lot of this metabolism occurs in your liver, and it’s “evolutionarily-designed function” is to detoxify (make unharmful) harmful things you might accidentally eat. So it’s sometimes nicknamed your body’s “detox center.” When you ingest something, your body wants to “screen” it, so it gets sent to the liver after being absorbed into the bloodstream in your intestines. The blood cycles through about once per minute. So even though the liver can’t modify it all the first time, it gets more and more chances. _ A lot of the metabolism that occurs is done by a family of protein enzymes (reaction mediators) called Cytochrome P450s (CYPs). CYPs are responsible for metabolizing ~75% of drugs on the market, but this work is split up among 57 family members. These guys are “heme-containing monooxygenases” -> like hemoglobin, they use heme as a cofactor to hold onto an iron atom. This iron atom then helps transfer oxygens onto things one (mono) at a time, “oxidizing them” PART 2
