Scientists in Britain have used gene-editing techniques to stop bird flu spreading in chicken cells grown in a lab - a key step towards making genetically-altered chickens that could halt a human flu pandemic. • Bird flu viruses currently spread swiftly in wild birds and poultry, and can at times jump into humans. Global health and infectious disease specialists cite as one of their greatest concerns the threat of a human flu pandemic caused by a bird flu strain that makes such a jump and mutates into a deadly and airborne form that can pass easily between people. • In the latest study, by editing out a section of chicken DNA inside the lab-grown cells, researchers from Imperial College London and the University of Edinburgh's Roslin Institute prevented the bird flu virus from taking hold in the cells and replicating. • "This is an important advance that suggests we may be able to use gene-editing techniques to produce chickens that are resistant to bird flu," Mike McGrew, who co-led the research, said in a statement. • In the further work, the team hopes to use CRISPR to remove a section of the birds' DNA responsible for producing a protein called ANP32, on which all flu viruses depend to infect a host. • Wendy Barclay, professor and chair in influenza virology at Imperial who worked with McGrew, says the idea behind developing gene-edited flu-resistant chickens is to be able "to stop the next flu pandemic at its source". • And she said work so far was showing promise: "We have identified the smallest possible genetic change we can make to chickens that can help to stop the virus taking hold." Cas13, an enzyme that bacteria use to fight off viruses, works by targeting not just the virus, but also the bacterium itself. It sends the bacterium into a dormant state and makes it an inhospitable place for viruses to reproduce. • That protects bacteria from mutated viruses that slip past other immune defenses, researchers report May 29, 2019, in the journal Nature. • Disarming or killing host cells is a common immune system strategy, but the new work is the first to demonstrate it for bacterial CRISPR defense, says molecular biologist Luciano Marraffini, a Howard Hughes Medical Institute investigator at the Rockefeller University. • "Cas13 has already become a very powerful diagnostic tool," says study coauthor Alexander Meeske, also at Rockefeller. Researchers can use it to quickly identify viruses in patients' blood, even those present at very low levels. "What we didn't know," he says, "is how Cas13's behavior affects bacterial immunity." • Now, he and his colleagues have found that Cas13's seemingly random snips are a valuable bacterial defense tool. While most Cas enzymes protect bacteria by stopping viruses from reproducing, Cas13 disables the bacterial host itself. • That's important because viruses can easily evade CRISPR systems—just a single genetic mutation in the region that the Cas enzyme targets can make the virus invisible to the immune system. A team of researchers from Tokyo University of Science (TUS), Meiji University, and Tokyo University of Agriculture and Technology, has recently established a series of novel strategies to increase the efficiency of targeted gene disruption and new gene "introduction" using the CRISPR-Cas9 system. • These strategies include quicker (single-step) gene introduction, use of small homologous sequences, and bypassing of certain prerequisite host DNA "patterns" and host component modification. • The team led by Takayuki Arazoe and Shigeru Kuwata has devised simple and quick techniques for gene editing (target gene disruption, sequence substitution, and re-introduction of desired genes) using CRISPR-Cas9 in the rice blast fungus Pyricularia (Magnaporthe) oryzae, a type of filamentous fungus. • Spurred on by encouraging results, the researchers surmise, "Plants and their pathogens are still coevolving in nature. Exploiting the mutation mechanisms of model pathogenic fungi as a genome editing technique might lead to the development of further novel techniques in genetic engineering." • "Rice blast fungus is an important pathogen that causes destructive disease of rice, which is the staple food of the country. The CRISPR-Cas9-based genome editing technique developed in our study can speed up molecular biological research on this pathogen, ultimately contributing to stable food supply and plant-based food safety." • "Also, this technique is applicable to other filamentous fungi widely used in industry--especially in the bioprocessing, food, and fermentation industries," the researchers said.
