This movie shows a molecular dynamics simulation of the protein MdmX in complex with a small molecule inhibitor, 298, otherwise known as . 4-[[(4S,5R)-5-(4-chlorophenyl)-4-(3-methoxyphenyl)- 2-(4-methoxy-2-propan-2-yloxy-phenyl)-4,5- dihydroimidazol-1-yl]carbonyl]piperazin-2-one . The inhibitor is shown as light, medium and dark pink spheres, and the MdmX protein is shown as red, blue, white, purple and yellow spheres. The colored spheres represent the individual atoms: White is hydrogen, blue carbon, red oxygen, purple nitrogen, and yellow sulfur. . The protein MdmX normally regulates the tumor suppressor protein, p53, by binding to it and preventing it from its functions in halting the cell cycle, repairing damaged DNA and inducing programmed cell death. . In some cancers, p53 activity is reduced by overexpression of MdmX, and this allows tumors to escape the p53-triggered cell death. To help restore p53 activity in these types of cancers, research has focused on finding inhibitors of MdmX. . The compound 298 was found in a search for small molecules that inhibit the MdmX-p53 interaction. Even though 298 binds to MdmX relatively weakly, it is able to overcome the inhibition of p53 in retinoblastoma cell lines where MdmX is overexpressed.
